The major structural differences between the two are that the two basic amino acid clusters in bipartite NLSs are separated by a relatively short spacer sequence (hence bipartite - 2 parts), while monopartite NLSs are not. The first NLS to be discovered was the sequence PKKKRKV in the SV40 Large T-antigen (a monopartite NLS). The NLS of nucleoplasmin, KR[PAATKKAGQA]KKKK, is the prototype of the ubiquitous bipartite signal: two clusters of basic amino acids, separated by a spacer of about 10 amino acids. === Relative and absolute quantification === Mass spectrometry is not inherently quantitative because of differences in the ionization efficiency and/or detectability of the many peptides in a given sample, which has sparked the development of methods to determine relative and absolute abundance of proteins in samples. The intensity of a peak in a mass spectrum is not a good indicator of the amount of the analyte in the sample, although differences in peak intensity of the same analyte between multiple samples accurately reflect relative differences in its abundance.

== Safety considerations == Inhalation of crystalline silica harms the lungs, causing silicosis. Amorphous silica is considered to have low toxicity, but prolonged inhalation causes lung changes. Diatomaceous earth is mainly amorphous silica but contains some crystalline silica, especially in the saltwater forms. In a 1978 study of workers, those exposed to natural diatomaceous earth for over five years had no significant lung changes while 40% of those exposed to the calcined form had developed pneumoconiosis. Today's standard diatomaceous earth formulations are safer to use, as they are predominantly made of amorphous silica and contain little or no crystalline silica. The crystalline silica content of diatomaceous earth is regulated in the United States by the Occupational Safety and Health Administration (OSHA). There are guidelines from the National Institute for Occupational Safety and Health that set maximum amounts allowable in the product (1%) and in the air near the breathing zone of workers, with a recommended exposure limit at 6 mg/m3 over an 8-hour workday. OSHA has set a permissible exposure limit for diatomaceous earth as 20 mppcf (80 mg/m3/%SiO2). At levels of 3,000 mg/m3, diatomaceous earth is immediately dangerous to life and health. In the 1930s, workers with long-term occupational exposure in the cristobalite diatomaceous earth industry who were exposed to high levels of airborne crystalline silica over decades were found to have an increased risk of silicosis.

== Prison escape and murder == On October 12, 2017, four inmates detained at the Pasquotank Correctional Institution attempted to conduct a prison escape, which led to the murders of four prison staff members and many others injured. On that day at 3pm, at the prison's sewing plant, the four perpetrators set a fire as a diversion, and using tools gathered from the prison's sewing factory, the four inmates attacked several prison staff members and their fellow prisoners by stabbing and beating them, before their escape attempt failed. As a result of the attack, two prison staff members, 35-year-old Corrections Officer Justin Smith and 50-year-old correction enterprises manager Veronica Darden, were mortally wounded and died on the day of the incident. Another four prisoners and ten prison staff members were injured during the attack, with three of the prison employees being hospitalized while in critical condition. As a result of the attack, the prison compound and nearby schools were placed on lockdown while searches were conducted for possible prison escapees, before it was confirmed that none of the prisoners had successfully escaped. At the time of the prison attack, 729 inmates were detained in Pasquotank Correctional Institution. On October 30, 2017, 49-year-old Correctional Officer Wendy Shannon succumbed to her injuries and died in the hospital, therefore becoming the third fatality of the prison attack.
=== Biogeography and environmental gradients === Ectomycorrhizal fungi are found throughout boreal, temperate and tropical ecosystems, primarily among the dominant woody-plant-producing families. Many of the fungal families common in temperate forests (e.g. Russulaceae, Boletaceae, Thelephoraceae) are also widespread in the Southern Hemisphere and tropical dipterocarp forests: although the plant families are quite different in temperate and tropical forests, the ectomycorrhizal fungi are fairly similar. The types of EcM fungi are affected by soil types both in the field and in the lab. For most types of plants and animals, species diversity increases towards the equator. This is called the latitudinal gradient of diversity (LGD). In contrast, there is evidence that EcM fungi may be at maximum diversity in the temperate zone. If this is the case, it might be explained by one or more of the following hypotheses: 1) EcM fungi may have evolved at higher latitudes with Pinaceae hosts, and be less able to compete in tropical climates; 2) the plants EcMs use as hosts might be more diverse in temperate conditions, and the structure of the soil in temperate regions may allow for higher niche differentiation and species accumulation; and 3) tropical EcM hosts are spread out more sparsely in small isolated forest islands that may reduce the population sizes and diversity of EcM fungi.
